The sensitivity and specificity of Next Generation Sequencing make it the ultimate quality control check for the Oligos used throughout it's workflows.

 

Biolegio's NGS range has been designed specifically for the synthesis of full-length adapter and indexing primers with dedicated production pipelines designed to produce the highest standard of DNA Oligo synthesis, with ultra-low cross-contamination of co-synthesised Oligos. 

 

In comparison with our standard Oligo production:

 

  • We utilse custom built, proprietary synthesis platforms
    • Designed in house by our Team, and their collective decades of knowledge on Oligo synthesis

 

  • Carefully designed Protocols for:
    • Handling, Post Synthesis Processing, dispensing, and annealing.

 

We ensure that every aspect of the production workflow has been considered and addressed to deliver high quality and performance NGS Indexing Oligos with cross-contamination levels of co-synthesised adapters and primer pairs at the lowest levels possible.

 

NGS QC Validated Production

 

Our NGS Synthesis production pipeline is process validated by the routine performance of an NGS based Quality Control assay, which demonstrates co-synthesised NGS Oligos within our NGS Grade pipeline maintain cross-contamination levels below 0.1% and as low as 0.01%.

 

NGS-S or NGS-P ?

NGS Desalted with NGS Grade QC checks

 

NGS-S is our standard NGS Grade Oligo synthesis service available and is recommended for customers for looking to order custom indexing primer pairs, for addition of Adapter sequences via PCR, or Full-Length ligation adapters where applications are not dealing with low sample input (<100ng).

NGS-P

 

NGS HPLC purification, Standard NGS QC plus an additional LC-MS check

 

NGS-P is recommended as the primary option for custom Full-Length ligation adapters

 

This is due to the length of full length adapter sequences and the inherent potential in solid support Oligo synthesis chemistry that the percentage of full length Oligo product will decrease as sequence length increases (see our Knowledge Base for more info).

 

HPLC purification will remove N-X product which may increase Adapter dimer levels if not removed and reduce library yield. This is crucial for low input samples where the percentage of truncated adapters to Sample will have more impact.

 

In order to maintain low cross-contamination risk, we not only observe more stringent handling procedures compared to our standard HPLC service, but also ensure that we use unique HPLC columns for every Oligo, Adapter or Index Pair purified.
